For MVP/PVM, Sera - painkiller is epic. You know how painkillers in real life make you feel like nothing can hurt you? Well it's like that in RO too, only here, that's a pretty accurate perception. Take 1's from most normal mobs with only basic gear. Watch waterballs miss. Etc. It's just insane. 

 

For WoE/MVP, one can argue for Dieter. It doesn't make you immortal like Sera does, but pyro gives +200 atk, and in WoE, Volcanic Ash is a brutal debuff (it's particularly effective for locking down snapping suras (even ones with packet-snap) by making snap fail.

 

Amistr is the best base type because it's the only one that helps the owner (with bulwark). All the others don't have anything that makes the owner more powerful. Vani levels fastest, though, and you need a vani if you plan to pot seriously with Prepare Potion (the alchemist skill) because Instruction Change adds 1% per level to potting rate.
