Hi, I'm currently in my last quest for Rune Knight (the quest where you can only carry your weapon and 400 weight). I have 70 dex but I always have trouble hitting with my skills. Do I just use the Rune they provide or how do I kill it? Are there really 3 monster to kill?

Use spear dynamo level 5 and you will have enough hit.

And do I use spiral clash to kill them?

Just spam Pertz rune to kill them. If you are running out of runes, then you use Crashing spiral.
