Doesnt AB bypass DEF already in its formula?

No. AB doesn't bypass HardDef and HardMdef. Def and Mdef subtract damage directly.

And PDM's Def and Mdef bypassing work on AB too.

BTW because of the formula. Increased damage is very negligible, around 1 or 2 digit depends on target Def and Mdef.

How do you even test this when the variance in damage is much higher than the def?

Simple. Test with bare hand give no damage variance.
Abusive robe is a good choice for this test. Test subject is high def demihuman or demon mobs such as Ygnigem or Seyren.

no wonder my AB does so much more dmg with the Devil's Hand. I only wanted to wear it for the fixed cast reduction and the uninterruptible skill cast, but was surprised to see it does about 100K more dmg on some bosses.

I guess that's your placebo effect '-' . Even with 200-300 Def. 100% bypassing should increase damage not more than 100 per hit.
And depends on how Devil's Hand Def bypassing works too.
- If it's similar to SS card. Its effect doesn't work on AB dmg at all.
- If it's similar to Abusive robe or PDM. It works. But as I said. Increased damage is very negligible.
