What are the quickest ways to level the homunculus' owner to get to genetic in order to turn it into a homunculus s (Without help)?
With my first biochemist I did:
Spores with level 6 archer merc
Ant eggs using cart revo + level 1/2/3/4 spear merc (650 ant eggs killed in 30 mins)
Semi afk Ant eggs to get enough job levels to summon my 99 sheep
Overlook 1/2/3/4/5 with sheep + level 5/6 archer merc killing
Brasilis 1/2 with level 6 archer merc + sheep + cart revo
2nd seal + new brasilis quest + most of the 91-99 eden board
Redo half of the 91-99 eden board to get job 70
Is there anything I missed or should have done differently that would have made it faster (I have 6 more level 99 evolved homunculus that need their owners to level).

aside from taking advantage of Gramps when available I think you hit everything.

After you hit base lvl 70-75 you could try the ongoing Gramps turn in quests. There's usually a quest for characters above level 70 or above 90 or something. They are great for AFK alchemists' job exp Just take your alchemist to the TI field and let the homu do the killing -> since after the balance patch the homun kills count too (which is great). Unfortunately it might take a long time to complete the turn ins if you are afk if the spawn rates are low.
Also the Eden group quest for green petites (hunt 200 green petites for food) is pretty nice.
Magma dungeon 1 or 2 if you have a party and Magmarings give good exp after you are level 96.

I did Eden quests and Brasilis Dungeon till 99/70...

cart revo earth petites or magmarings lv 95+ is how I did it, plus gramps turnins of course. And yes, brasilis is an amazing little undiscovered gem for most people. Just grab some water armor and go to town.
